Analysis on Effect Factors of Hydrolysis Efficiency of Industrial Cellulases

Abstract: The factors affecting the efficiency of enzymatic hydrolysis by Accellerase 1000, and the glucose inhibition and deactivation property of several typical industrial cellulases including Accellerase 1000, Spezyme CP, and Novozyme 188 were investigated. The results showed that the hydrolysis efficiency was not significantly affected by the reactor geometry, while the solid content, the cellulase loading, the stirring rate and the temperature had great influence on the hydrolysis efficiency of Accellerase 1000. The activities of filter paper units (FPU) and cellubioase (CBU) of Accellerase 1000 decreased faster in the acetate buffer than Spezyme CP and Novozyme 188. With the existence of lignin particles, the deactivation rate of the filter paper units was decreased, while the deactivation rate of cellubioase activity was increased. The activities of Spezyme CP and Novozyme 188 were more stable than Accellerase 1000. Glucose showed an obvious inhibition effect on the cellulase enzymes.
